چکیده

A highly sensitive, fast, and stable conductometric enzyme biosensor for the determination of nitrate in water is described and validated in natural water samples. The nitrate biosensor is based on a methyl viologen mediator mixed with nitrate reductase (NR) from Aspergillus niger and Nafion® cation-exchange polymer dissolved in a plasticized PVC membrane deposited on the sensitive surface of interdigitated electrodes. The process parameters for the fabrication of the enzyme biosensor and various measuring conditions such as pH buffer concentration and temperature were investigated with regard to their effect on sensitivity, limit of detection, dynamic range and operational and storage stabilities. The sensitivity of the nitrate sensor was approximately 1.48 μS·L/mg, the detection limit was 1.2 μg/L, and linear calibration was in the range from 4×10–3 to 8 mg/L with an application domain from 4×10–3 to 50 mg/L. When stored in 20 mM phosphate buffer (pH 7.5) at 4°C, the sensor showed good stability over 2 months.

A Nitrite Biosensor Based on Co-immobilization of Nitrite Reductase and Viologen-modified Chitosan on a Glassy Carbon Electrode

An electrochemical nitrite biosensor based on co-immobilization of copper-containing nitrite reductase (Cu-NiR, from Rhodopseudomonas sphaeroides forma sp. denitrificans) and viologen-modified chitosan (CHIT-V) on a glassy carbon electrode (GCE) is presented. Electron transfer (ET) between a conventional GCE and immobilized Cu-NiR was mediated by the co-immobilized CHIT-V. Redox-active viologen...

Printed in Great Britain Sites and Specificity of the Reaction of Bipyridylium Compounds with Anaerobic Respiratory Enzymes of Escherichia coli EFFECTS OF PERMEABILITY BARRIERS IMPOSED BY THE CYTOPLASMIC MEMBRANE By ROBERT W. JONES and PETER B. GARLAND

The ability of the oxidized and singly reduced species of several bipyridylium cations to cross the cytoplasmic membrane of Escherichia coli was studied to locate the sites of reaction of the dyes with anaerobic respiratory enzymes. Benzyl Viologen radical crossed the membrane rapidly, whereas the oxidized species did not. The oxidized or radical species of Methyl Viologen, Morfamquat or Diquat...

Selenate reduction by Enterobacter cloacae SLD1a-1 is catalysed by a molybdenum-dependent membrane-bound enzyme that is distinct from the membrane-bound nitrate reductase.

Enterobacter cloacae SLD1a-1 is capable of reducing selenium oxyanions to elemental selenium under both aerobic and anaerobic conditions. In this study the enzyme that catalyses the initial reduction of selenate (SeO4(2-)) to selenite (SeO3(2-)) has been localised to isolated cytoplasmic membrane fractions. Experiments with intact cells have shown that the putative selenate reductase can accept...

Properties of a thermostable nitrate reductase from the hyperthermophilic archaeon Pyrobaculum aerophilum.

The nitrate reductase of the hyperthermophilic archaeon Pyrobaculum aerophilum was purified 137-fold from the cytoplasmic membrane. Based on sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis, the enzyme complex consists of three subunits with apparent molecular weights of 130,000, 52,000, and 32,000. The enzyme contained molybdenum (0.8-mol/mol complex), iron (15.4-mol/mol comp...
